The application itself doesn't support them. It is a job for the plugins to implement it. If you are interested in .mp3, then the in_mpg123 plugin is what you need.
So what you're saying is, "there's no nice, clean, simple way of scanning, applying, and reading replaygain tags with winamp"?
If that's the case then ...

Winamp lacks many "nice, clean and simple solutions" for quite a few things.

Foobar2000 "invented" and spearheaded the "clean and cross-format"-replaygain approach. Other apps are still lagging behind but are *slowly* catching up after like........ 2 years. But they *are* catching up at least ;)
If you are after player which is visually "wimamp-alike", then you may want to check out XMPlay ( www.un4seen.com ). IIRC it does support replaygain (not sure if that includes scanning). It also supports sonique-visualizations and winamp-inputplugins. Since about a year, XMPlay has begun to implement some of the features which until then were mostly fb2k-only.... correct unicode-support, *simple* titleformatting and.... replaygain. The rapid development of xmplay has gone mostly unnoticed here at ha.org, but this player has improved alot in flexibility in the recent year.
